Dark field microscope is mainly used for observation of objects whose biological structure is related to refractive index changes, such as diatoms, radiation insects, bacteria and other single-cell organisms with regular structures, as well as linear structures such as flagella, fiber inside cells. Besides, dark field biological microscope can observe particles below the resolution limits of objective lens.
